Diverse management techniques have been used to mitigate conflicts between humans and double-crested cormorants (Phalacrocorax auritus) including harassment methods supplemented by lethal take. In this study we evaluated impacts of programs to harass spring migrating cormorants on the walleye (Sander vitreus) fishery in Brevoort Lake and the yellow perch (Perca flavescens) and walleye fisheries at Drummond Island. Cormorant foraging declined significantly (p < 0.05) at both locations subsequent to initiation of harassment programs. Overall harassment deterred 90% of cormorant foraging attempts while taking less than 6% lethally on average at each site. Yellow perch were a predominate prey item in number and biomass at both locations. Walleye made up a small proportion of the diet at both locations. However, both walleye and yellow perch abundance increased significantly (p < 0.05) at Drummond Island. Walleye abundance at age 3 increased to record levels in 2008 following 3 years of cormorant management at Brevoort Lake. The estimated cormorant consumption of age-1 walleye in the absence of management at Brevoort Lake during 2005 would account for 55% of the record 2006 age-1 walleye population. These results support the hypothesis that cormorant predation on spawning aggregations of sportfish was a significant mortality factor and cormorant management reduced sportfish mortality and increased abundance at both locations. Continuation of harassment programs and fishery assessments will determine whether improvement of targeted sport fisheries through control of spring migrating cormorants is sustainable.  相似文献

An increase in the proportion of unsaturated fatty acids in milk is considered desirable for human health. A prerequisite for the manipulation of milk fat composition is a co-ordinated understanding of the complex interactions in its biosynthesis. It has been suggested that an increase in the expression of mammary stearoyl-CoA-desaturase (SCD) would enrich mono-unsaturated fatty acids in milk, and therefore improve its nutritional properties. To investigate the potential effects of changes in expression of mammary enzymes and substrate availability on milk fat composition, we constructed, parameterized and evaluated a mechanistic mathematical model of fatty acid biosynthesis and milk-fat triglyceride assembly. The objective was to describe changes in the amount and composition of milk fat produced by bovine mammary cells due to changes in nutrition. Using the model we found that a 50% up-regulation in SCD activity increased the molar fraction of milk triglyceride 18:1 from 0.30 to 0.33 and 16:1 from 0.04 to 0.06. Up-regulation of SCD therefore did not appear to be the optimal method for increasing the content of unsaturated fatty acids in milk fat. The model was also used to determine the likely rate-limiting processes for the incorporation of unsaturated fatty acids into milk fat. Halving the concentration of glycerol 3-phosphate increased the molar fraction of milk triglyceride 18:1 from 0.30 to 0.35 and decreased the molar fraction of milk triglyceride 16:0 from 0.30 to 0.22. This achieved the desirable outcome of producing more unsaturated low-fat milk. Our model also predicted that a K232A mutation in the bovine mammary DGAT1 gene that is linked with an increase in milk fat yield would be consistent with a 120% increase in the DGAT acylation rate and also would be associated with a decrease in milk mono-unsaturated fatty acids.  相似文献

BACKGROUND: A transgenic papaya line (TPY10‐4) that is resistant to both papaya ringspot virus (PRSV) and papaya leaf distortion mosaic virus (PLDMV) has been developed in Taiwan. This study investigated the immunomodulatory properties of transgenic TPY10‐4 and its native (TCK) papaya fruits using an ovalbumin (OVA)‐sensitised mouse model. Both green and ripe papaya fruits at low (0.2 g powder kg?1 body weight (BW)) and high (1.6 g powder kg?1 BW) doses were administered to experimental mice by intragastric gavage for 5 weeks. Changes in serum total immunoglobulin A (IgA), IgE, IgG and IgM levels, OVA‐specific IgE, IgG1 and IgG2a titres and Th1/Th2 cytokine secretions using splenocytes were determined. RESULTS: Transgenic TPY10‐4 or native TCK papaya fruit supplementation did not significantly affect body, visceral organ and relative tissue weights, total IgE antibody levels, OVA‐specific IgE and IgG1 antibody titres or OVA‐stimulated interferon‐γ (IFN‐γ), interleukin‐2 (IL‐2), IL‐4, IL‐5 and IL‐10 secretions using splenocytes. However, transgenic papaya fruits markedly increased serum total IgM levels. CONCLUSION: This study suggests that transgenic TPY10‐4 papaya fruits do not increase the allergenic potential of OVA by oral administration but may have a protective immunity via increasing the serum total IgM level. Our objective was to assess the effects of feeding negative dietary cation-anion difference (DCAD) prepartum diets on milk production, reproductive performance, and culling. Cows from 4 commercial farms in Ontario, Canada were enrolled in a pen-level controlled trial from November 2017 to April 2019. Close-up pens (1 per farm) with cows 3 wk before calving were randomly assigned to a negative DCAD (TRT; ?108 mEq/kg of dry matter; target urine pH 6.0–6.5) or a control diet (CON; +105 mEq/kg of dry matter with a placebo supplement). Each pen was fed TRT or CON for 3 mo (1 period), and then switched to the other treatment for the next period (4 periods per farm). Data from 15 experimental units (8 pen treatments in TRT and 7 in CON), with a total of 1,086 observational units (cows), were included. The effect of treatment on milk yield at the first 3 milk recording tests of lactation was assessed with linear regression models accounting for repeated measures. The risk of pregnancy at first artificial insemination and culling by 30, 60, and 305 d in milk (DIM) were analyzed with logistic regression models, and effects on time to first AI, pregnancy, and culling were assessed with Cox proportional hazards models. All models included treatment, parity, and their interactions, accounting for pen-level randomization and clustering of animals within farm with random effects, giving 10 degrees of freedom for treatment effects. Multiparous cows fed TRT produced more milk at the first (42.0 vs. 38.8 ± 1.2 kg/d) and second (44.2 vs. 41.7 ± 1.3 kg/d) milk tests. However, multiparous cows fed TRT tended to have 0.2 percentage units less milk fat content at these tests. Although multiparous cows fed TRT tended to have greater energy-corrected milk at the first test (least squares means ± standard error: TRT = 46.1 ± 0.9 vs. CON = 43.8 ± 1 kg/d), there were no differences observed in energy-corrected milk at the second or third tests. In primiparous cows, there was no effect of treatment on milk production. Multiparous cows fed TRT had greater pregnancy to first insemination (TRT = 42 ± 3 vs. CON = 32 ± 4%) and tended to have shorter time to pregnancy [hazard ratio (HR) = 1.20; 95% CI: 0.96–1.49]. In primiparous cows fed TRT, time to pregnancy was increased (HR = 0.76; 95% CI: 0.59–0.99). Culling by 30 DIM tended to be less in TRT (3.3 ± 1.1%) than CON (5.5 ± 1.8%). No effect of treatment on culling by 305 DIM was detected in primiparous cows, but in multiparous cows, the TRT diets decreased the odds of culling (21.3 ± 1.9 vs. 31.7 ± 2.8%) and daily risk of culling to 305 DIM (HR = 0.64; 95% CI: 0.46 to 0.89). Under commercial herd conditions, prepartum negative DCAD diets improved milk production and reproductive performance, and reduced culling risk in multiparous cows. In primiparous cows, TRT diets had no effect on milk yield or culling, but increased the time to pregnancy. Our results suggest that negative DCAD diets should be targeted to multiparous cows.  相似文献

Cold-loving or psychrophilic organisms are widely distributed in nature as a large part of the earth's surface is at temperatures around 0 degrees C. To maintain metabolic rates and to prosper in cold environments, these extremophilic organisms have developed a vast array of adaptations. One main adaptive strategy developed in order to cope with the reduction of chemical reaction rates induced by low temperatures is the synthesis of cold-adapted or psychrophilic enzymes. These enzymes are characterized by a high catalytic activity at low temperatures associated with a low thermal stability. A study of protein adaptation strategies suggests that the high activity of psychrophilic enzymes could be achieved by the destabilization of the active site, allowing the catalytic center to be more flexible at low temperatures, whereas other protein regions may be destabilized or as rigid as their mesophilic counterparts. Due to these particular properties, psychrophilic enzymes offer a high potential not only for fundamental research but also for biotechnological applications.  相似文献

A new accelerated weathering protocol has been developed which closely replicates the performance of automotive and aerospace coating systems exposed in South Florida. IR spectroscopy was used to verify that the chemical composition changes that occurred during accelerated weathering in devices with a glass filter that produced a high fidelity reproduction of sunlight’s UV spectrum matched those that occurred during natural weathering. Gravimetric water absorption measurements were used to tune the volume of water absorption during accelerated weathering to match that which occurred during natural weathering in South Florida. The frequency of water exposure was then scaled to the appropriate UV dose. A variety of coating systems were used to verify the correlation between the physical failures observed in the accelerated weathering protocol and natural weathering in South Florida. The new accelerated weathering protocol correctly reproduced gloss loss, delamination, cracking, blistering, and good performance in a variety of diverse coating systems. For automotive basecoat/clearcoat paint systems, the new weathering protocol shows significant acceleration over both Florida and previous accelerated weathering tests. For monocoat aerospace systems, the new weathering protocol showed less acceleration than for automotive coatings, but was still an improvement over previous accelerated tests and was faster than Florida exposure.  相似文献

However, their inclusion forces one to address the issues of completeness and consistency that naturally arise from this organizational role. The outcome of this work is a knowledge-based associate ergonomic tool that assists in determining visibility of stationary small objects critical to the work environment. The Luchlesh and Moss detection solid, a design handbook guideline, accounts for non-search visibility in terms of three fundamental variables — visual angle, brightness and contrast. To reduce the difficulty of using the solid in its current form, this effort first quantified the empirical relationships of the solid. The intelligent system building tool, Exsys, was then employed: (1) to convert the results into rules for a knowledge base, and (2) to generate a tool called the Visual Detectability Analysis Associate. An efficient, convenient and easy to use ergonomic design tool is now available that determines visibility of small stationary objects in work environments where viewing time may be limited, but visual search is not required. In current use by a company to solve visibility problems encountered in system design, the VDAA tool has demonstrated beneficial savings in both time and effort.  相似文献
